I want to give students a grade for an assignment, but I don't want this grade to be incorporated into the student's overall mean average for the course - is there a way of doing this?

If you're using weighted mean you can leave the weight of the assignment 0. If you don't use weight mean put all assignments that count towards the course grade in a category and set its aggregation method to whatever works for you. Set the course category aggregation to weighted mean, the weight of the above category to 1.0 and the weight of the extra assignment to 0. See illustration below.
- Course category (aggregation: weighted mean)
- Included assignments category (aggregation: simple mean; weight: 1)
- Assignment 1
- Assignment 2
- Assignment 3
- Extra assignment (weight: 0)
- Included assignments category (aggregation: simple mean; weight: 1)

Should work. This approach uses only basic features of the gradebook. Should be easy enough to try.
