Can anyone assist, a few colleagues have completed a Moodle course/ summative assessment, however, their grades aren't reading through to the grade book.
Can anyone guide me on what could possibly be the issue or if there's a setting that I need to enable?

The activity is graded automatically. It's made of a few readings, material etc and an assessment.

In reviewing the screen capture you provided the student only completed 2 out of 5 activities to set the grade. They viewed the summative assessment and made an attempt. However, they did not receive a grade, receive a passing grade or complete all attempts.
Are you sure that you want to require all of these options for the activity? If you must keep the requirements each student will need to complete all of the requirements to receive a grade.
