Once a password policy is enabled on a site that didn't previously have one, will current users be asked to change their passwords to reflect the policy, or will the policy only affect new user accounts?

It will only affect new users. However, existing users will need to follow the requirements on a password update. You could force that for all users...

Go to bulk user options, select all users, force password change...
