Is it possible to require an enrolment key for a course for say guests, but allow students to get straight in via internal enrolment?

Add an enrollment key and in the drop box for guest access select allow guests who have the key. Enroll the student via whatever method you are using and they shouldn't need to know the key-they should just get in. guests should be prompted to enter the key. Check out this page for more info

One caveat: I have not been able to get this to work correctly on my site, but I know others have had success with it. My need went away so I just didn't explore the idea further...
