I have seen that message many times and it almost always means what it says. $CFG->dataroot is pointing to somewhere that either does not exist or the web server user (apache?) does not have permission to see that folder. Do you know what has changed recently on your system?

I have a similar problem. I was using normally yesterday (April 24) and today I get the following message: "Error reading from database".
