I defined all the subject's questions as "Top for System" maintaining unique "Question Name".  Maintaining a single question bank.

I am planning to create a quiz of 100 questions for 400 marks in which 1-25 questions belongs to one subject 26-50 subject 2; 51-75 subject 3; 76-100 subject 4. Each correct answer gets 4 Marks, Each wrong answer gets 1 Negative mark, Unattempt gets 0 mark. 

Is there any possible way of doing this. with or without plugins. 

And I want it to be active at defined time.

Your use case would depend on which Question Types (QT) you will be using.
If you are using True/False this is simple. If you are using Multiple Choice Questions (MCQ) this will take some math skills to allocate the correct point values to each question. Map this out to achieve your desired results offline before your start creating questions in Moodle.

This is perfectly doable with standard Moodle features. Just set it up. See https://docs.moodle.org/en/Building_Quiz if you need documetation.

(Also, while learning, perhaps practice creating an 8-question quiz, with a total score of 32, with 2 questions from each category. That way you can learn and gain confidence more quickly.)
