I can see this setting in the Self enrolment, is there this possibility when the students are enrolled to a course manually?

Good question, and I think that this question has been asked before. I don't think that this message (the one on this setup page) gets emailed for manual enrollments. On my moodle, this email is sent as a way of letting the student know that they completed the self-enrollment. I use regular email (my client email) to let students know when I manually enroll them, and somehow, this has never bothered me.
Maybe someone else will know more than I do. Let's see.
