This is not possible in Moodle within the video. What I have seen done is to use a Lesson and divide the lecture up into parts. The questions can then come at a break between parts and then move on to the next page and have another part of the lecture video. This will put the scores in the gradebook and actually gives students a easy way to go back and review the lecture part they need to quite easily latter in the course.
