A three column theme means blocks on left and right side and in the middle of the three columns the content of your page (resources, activities) - therefore the middle part technically is not a block and you can´t add a block like 'Slideshow' here. Or do I misunderstand you? What the Moodle version and the theme you are using?

Anyway, as I said above, it isn´t possible using any block in the middle. What you can do is using Activity Module: Image Slideshow or Resource Type: Lightbox Gallery Resource (but both without random assignment)...
